Project 3 (3)

PRESENT PROJECTS

  1. Define novel proteins that interact with IGF-I receptor or insulin receptor by two-hybrid assay, or co-immunoprecipitation proteomics analysis, and elucidate physiological roles of these proteins
    • Insulin receptor or insulin-like growth factor I receptor-associated proteins (InsRAPs and IGFRAPs)
    • 14-3-3, SOCS proteins, Grb10, PI 3-kinase

  2. Define novel proteins that interact with IRS family proteins by two-hybrid assay and co-immunoprecipitation proteomics analysis
    • Signal-modulating proteins; 14-3-3
    • Endosome sorting proteins; AP-1 and AP-2 components, importin
    • Transcription regulator proteins; TYSLP, BMI
    • Phosphorylation modulators; 53BP2, DGK, IRSAL
    • Ubiquitination and deubiquitination enzymes; Nedd4, Mdm2, and UPSs

    • Endosomal localization of IRS-1 and IRS-2
    • Mitochondrial localization of IRS-1 and IRS-2
    • Nuclear localization of IRS-3

  4. Characterize intramolecular modification of IRS family proteins under various conditions.

  5. Screening of small molecules for capacity to modulate binding between IRS and IRS-associated proteins, IGF-I receptor and associated proteins, and insulin receptor and associated proteins
    • Small compounds in chemical library in Riken
